【适应症/功能主治】 精神分裂症。

【规格型号】 4mg*10s

【用法用量】 一般成人的初始剂量维每次4mg,每日两次,餐后服用。根据患者的年龄及症状,可适当增减剂量,维持剂量为每日8-16mg,每日剂量不应超过24mg。详情见说明书。

【注意事项】 
请仔细阅读说明书并遵医嘱使用。

【儿童用药】 本品在18岁以下的儿科人群中的安全有效性尚未确立。

【老年患者用药】 老年患者通常生理功能较弱,容易出现锥体外系症状,因此应密切观察患者状态,慎重用药。

【孕妇及哺乳期妇女用药】 (1) 对于孕妇或可能怀孕的妇女,只有在服用本品的益处大于危险的情况下才能用药。(本品在妊娠期间给药的安全性尚未确立。有报告称,于妊娠后期给予抗精神病类药品时,出现了新生儿喂养障碍、嗜睡、呼吸系统疾病、震颤、 张力减低、易激惹等戒断症状及锥体外系症状。) (2) 哺乳期妇女在服用本品期间应停止哺乳(动物试验(大鼠)结果表明,本品可从乳汁分泌)。

【药物过量】 目前尚缺乏药物过量的临床资料。

【药理毒理】 布南色林为多巴胺受体及5-羟色胺受体拮抗剂。体外受体结合试验结果显示,布南色林对于多巴胺D2受体亚型(D2、D3)及5-HT2A受体表现出亲和性。其主要代谢产物N-脱乙基体对于多巴胺D2受体亚型(D2、D3)及5-HT2A受体也表现出亲和性,但对多巴胺D2受体的亲和性仅为布南色林的1/10左右。另外,N-脱乙基体对于5-HT2c受体及5-HT6受体也表现出亲和性。但布南色林及N-脱乙基体对于肾上腺素α1、组胺H1、蕈毒碱M1等受体的亲和性较低。 毒理研究 遗传毒性: 布南色林Ames试验、CHL/IU细胞染色体异常试验及小鼠微核试验结果均为阴性。 生殖毒性: 大鼠及家兔生殖发育毒性试验结果未见布南色林具有致畸性。 致癌性: 雌雄大鼠连续104周,每日1次经口给予布南色林0.1、0.3及1mg/kg,雌性动物的高剂量(1mg/kg/日)组乳腺瘤发生率升高。另外,雌雄小鼠连续104周, 每日1次经口给予布南色林1、3及10mg/kg,雌性动物在所有剂量水平下乳腺瘤以及垂体瘤的发生率升高。   在大鼠以及小鼠中观察到的这些变化,为多巴胺D2受体阻断作用的药物对啮齿类动物进行给药时常见的变化,认为是由于催乳素分泌亢进所导致。 依赖性: 布南色林大鼠身体依赖性试验中未见身体依赖性的形成。猴胃内自身给药试验中,将布南色林换成替代介质后,4只动物中有1只出现与强化消除抗拒相似的、 一过性自身给药次数增加的现象。但在布南色林的自身给药期内,并未出现可判断为强化效应阳性的、自身给药次数增加的现象,可以认为布南色林导致人体产生药物依赖或者药物滥用的可能性较低。

Description

It regulates neurotransmitter in the brain (e.g. dopamine, serotonin) to soothe strong anxiety/tense nerve and stabilize the mood.
It is usually used to treat schizophrenia.

Effect/Efficacy

It regulates neurotransmitter in the brain (e.g. dopamine, serotonin) to soothe strong anxiety/tense nerve and stabilize the mood.
It is usually used to treat schizophrenia.

Usage/Dosage

In general, for adults, start with taking 1 tablet (4mg of the active ingredient) at a time, twice a day after meals. Then, increase the dose in incremental steps. Standard maintenance dose is 1-2 tablet (4-8mg) at a time, and twice daily after meals. The dosage may be adjusted according to the age or symptoms. Daily dose must not exceed 6 tablets (24mg). Strictly follow the instructions.
If you dose on empty stomach, this medicine may not work. Be sure to take the medicine after meals.
If you are ordered to use this medicine on regular schedule and you missed a dose, eat something and take as soon as possible. If it is almost time for the next dose, skip the missed dose and continue your regular dosing schedule. You should never take two doses at one time.
If you accidentally took more than your prescribed dose, consult with your doctor or pharmacist.
Do not stop taking this medicine unless your doctor instructs you to do so.

Cautions

If you have previously experienced any allergic reactions (itch, rash, etc.) to any medicines.
If the patient is in a coma.
If you are pregnant or breastfeeding.
If you are taking any other medicinal products. (Some medicines may interact to enhance or diminish medicinal effects. Beware of over-the-counter medicines and dietary supplements as well as other prescription medicines.)

This drug may cause drowsiness and disorders in: attentiveness, concentration, and reflex movement. Therefore do not drive a car or operate dangerous machinery while taking this medicine.
Please note that alcohol may intensify the effect of this medicine.
Do not take grapefruit juice and this medicine together as it may intensify the effect of this medicine.

Keep out the reach of children. Store away from direct sunlight, heat and moisture.
Discard the remainder. Do not store them. Ask your pharmacist how to discard.

Contraindication

The most commonly reported adverse reactions include Parkinson's syndrome (tremor in limbs, muscle rigidity, slow movement), akathisia (restlessness, walking around), insomnia, dyskinesia (involuntary movement of face, particularly around the mouth), drowsiness, anxiety, feeling irritated, irritability, rash, eczema and itch. If any of these symptoms occur, consult with your doctor or pharmacist.
The symptoms described below are rarely seen as initial symptoms of the adverse reactions indicated in brackets. If any of these symptoms occur, stop taking this medicine and see your doctor immediately.
sudden fever, muscle rigidity, hands/feet tremor [malignant syndrome]
involuntary movement of face, particularly around the mouth [tardive dyskinesia]
loss of appetite, nausea, vomiting, severe constipation [paralytic ileus]
loss of appetite, nausea, vomiting, convulsion [syndrome of inappropriate antidiuretic hormone secretion]
pain/stiffness/numbness in the muscles of the limbs [rhabdomyolysis]
The above symptoms do not describe all the adverse reactions to this medicine. Consult with your doctor or pharmacist if you notice any symptoms of concern other than those listed above.
